    GLEAM v3 : satellite-based land evaporation and root-zone soil moisture

    The Global Land Evaporation Amsterdam Model (GLEAM) is a set of algorithms dedicated to the estimation of terrestrial evaporation and root-zone soil moisture from satellite data. Ever since its development in 2011, the model has been regularly revised, aiming at the optimal incorporation of new satellite-observed geophysical variables, and improving the representation of physical processes. In this study, the next version of this model (v3) is presented. Key changes relative to the previous version include (1) a revised formulation of the evaporative stress, (2) an optimized drainage algorithm, and (3) a new soil moisture data assimilation system. GLEAM v3 is used to produce three new data sets of terrestrial evaporation and root-zone soil moisture, including a 36-year data set spanning 1980-2015, referred to as v3a (based on satellite-observed soil moisture, vegetation optical depth and snow-water equivalent, reanalysis air temperature and radiation, and a multi-source precipitation product), and two satellite-based data sets. The latter share most of their forcing, except for the vegetation optical depth and soil moisture, which are based on observations from different passive and active C-and L-band microwave sensors (European Space Agency Climate Change Initiative, ESA CCI) for the v3b data set (spanning 2003-2015) and observations from the Soil Moisture and Ocean Salinity (SMOS) satellite in the v3c data set (spanning 2011-2015). Here, these three data sets are described in detail, compared against analogous data sets generated using the previous version of GLEAM (v2), and validated against measurements from 91 eddy-covariance towers and 2325 soil moisture sensors across a broad range of ecosystems. Results indicate that the quality of the v3 soil moisture is consistently better than the one from v2: average correlations against in situ surface soil moisture measurements increase from 0.61 to 0.64 in the case of the v3a data set and the representation of soil moisture in the second layer improves as well, with correlations increasing from 0.47 to 0.53. Similar improvements are observed for the v3b and c data sets. Despite regional differences, the quality of the evaporation fluxes remains overall similar to the one obtained using the previous version of GLEAM, with average correlations against eddy-covariance measurements ranging between 0.78 and 0.81 for the different data sets. These global data sets of terrestrial evaporation and root-zone soil moisture are now openly available at www.GLEAM.eu and may be used for large-scale hydrological applications, climate studies, or research on land-atmosphere feedbacks

    Transductive Learning with String Kernels for Cross-Domain Text Classification

    For many text classification tasks, there is a major problem posed by the lack of labeled data in a target domain. Although classifiers for a target domain can be trained on labeled text data from a related source domain, the accuracy of such classifiers is usually lower in the cross-domain setting. Recently, string kernels have obtained state-of-the-art results in various text classification tasks such as native language identification or automatic essay scoring. Moreover, classifiers based on string kernels have been found to be robust to the distribution gap between different domains. In this paper, we formally describe an algorithm composed of two simple yet effective transductive learning approaches to further improve the results of string kernels in cross-domain settings. By adapting string kernels to the test set without using the ground-truth test labels, we report significantly better accuracy rates in cross-domain English polarity classification.Comment: Accepted at ICONIP 2018. arXiv admin note: substantial text overlap with arXiv:1808.0840

    Negative time delay for wave reflection from a one-dimensional semi-harmonic well

    It is reported that the phase time of particles which are reflected by a one-dimensional semi-harmonic well includes a time delay term which is negative for definite intervals of the incoming energy. In this interval, the absolute value of the negative time delay becomes larger as the incident energy becomes smaller.     Emulsifying properties of defatted rice bran concentrates enriched in fiber and proteins

    BACKGROUND: Rice bran (RB), a by-product of the rice milling industry, constitutes around 10% of the total weight of rough rice. The interest in the use of RB is centered on its nutritional quality, its low cost, and its extensive worldwide production. As RB is commonly used for oil extraction, the defatted rice bran (DRB) is obtained as a second by-product. The aim of this work was to obtain a defatted rice bran concentrate (DRBC), enriched in protein and fiber, from defatted rice bran flour (DRBF) and to determine its physicochemical and emulsifying properties. RESULTS: To obtain the DRBC, the starch was efficiently hydrolyzed (> 98%) with α-amylase and amyloglucosidase, with a concomitant increase in the proportions of crude protein (from 154.7 to 274.3 g kg−1) and total dietary fiber (from 276.1 to 492.3 g kg−1). Defatted rice bran concentrate exhibited a loss of protein solubility and increased surface hydrophobicity compared with DRBF. Defatted rice-bran concentrate dispersions with and without previous ultrasound treatment were prepared. The sonication led to an increase in the apparent viscosity. Emulsions were prepared with dispersions with and without previous ultrasound treatment and showed high stability in quiescent conditions over 28 days. However, the emulsions prepared with dispersions treated with ultrasound resulted in lower D4,3 values and higher elastic and viscous moduli. CONCLUSION: The rice bran concentrate can be used to obtain stable oil-in-water (O/W) emulsions, including both soluble and insoluble fractions, in acidic and neutral conditions.     Non-L\'evy mobility patterns of Mexican Me'Phaa peasants searching for fuelwood

    We measured mobility patterns that describe walking trajectories of individual Me'Phaa peasants searching and collecting fuelwood in the forests of "La Monta\~na de Guerrero" in Mexico. These one-day excursions typically follow a mixed pattern of nearly-constant steps when individuals displace from their homes towards potential collecting sites and a mixed pattern of steps of different lengths when actually searching for fallen wood in the forest. Displacements in the searching phase seem not to be compatible with L\'evy flights described by power-laws with optimal scaling exponents. These findings however can be interpreted in the light of deterministic searching on heavily degraded landscapes where the interaction of the individuals with their scarce environment produces alternative searching strategies than the expected L\'evy flights. These results have important implications for future management and restoration of degraded forests and the improvement of the ecological services they may provide to their inhabitants.Comment: 15 pages, 4 figures.     Severity and duration of allergic conjunctivitis: are they associated with severity and duration of allergic rhinitis and asthma?

The association of allergic conjunctivitis (AC) with rhinitis and/or asthma is poorly understood. The objective of this study was to apply the Consensus Document for Allergic Conjunctivitis (DECA) criteria for the classification of AC to a population of patients with AC to assess the association between the severity and duration of AC and rhinitis and/or asthma. Methods. Patients with ocular symptoms of AC who participated in the 'Alergológica 2015' study were included. The demographics, classification according to the DECA criteria, etiology, and comorbidities were evaluated by age groups (less or equal than 14 and greater than 14 years). Results. A total of 2,914 patients (age range, 1-90 years) were included in the "Alergológica 2015" study. Of these, 965 patients (33.1%) were diagnosed with AC (77.5% > 14 years). AC was classified as severe, moderate, or mild in 1.8%, 46.4%, and 51.8%, respectively; and as intermittent or persistent in 51.6% and 48.4% of the patients. AC alone occurred in 4% of patients. AC was mainly associated with rhinitis (88.4%), asthma (38.2%), food allergy (8.3%) and atopic dermatitis (3.5%). In allergic respiratory disease rhinitis preceded AC and asthma developed later. The severity and duration of AC was significantly associated with severity and duration of rhinitis (p less than 0.001 for both age groups) and asthma (p less than 0.001 only in adults). Conclusions. The application of the new DECA classification for AC reveals a direct relationship between AC, rhinitis and asthma respect to severity and duration. These relationships suggest that AC should be considered an integral part of the "one airway, one disease" hypothesis

    The Structural Architecture of an Infectious Mammalian Prion Using Electron Cryomicroscopy

    The structure of the infectious prion protein (PrPSc), which is responsible for Creutzfeldt-Jakob disease in humans and bovine spongiform encephalopathy, has escaped all attempts at elucidation due to its insolubility and propensity to aggregate. PrPSc replicates by converting the non-infectious, cellular prion protein (PrPC) into the misfolded, infectious conformer through an unknown mechanism. PrPSc and its N-terminally truncated variant, PrP 27-30, aggregate into amorphous aggregates, 2D crystals, and amyloid fibrils. The structure of these infectious conformers is essential to understanding prion replication and the development of structure-based therapeutic interventions. Here we used the repetitive organization inherent to GPI-anchorless PrP 27-30 amyloid fibrils to analyze their structure via electron cryomicroscopy. Fourier-transform analyses of averaged fibril segments indicate a repeating unit of 19.1 Å. 3D reconstructions of these fibrils revealed two distinct protofilaments, and, together with a molecular volume of 18,990 Å3, predicted the height of each PrP 27-30 molecule as ~17.7 Å. Together, the data indicate a four-rung β-solenoid structure as a key feature for the architecture of infectious mammalian prions. Furthermore, they allow to formulate a molecular mechanism for the replication of prions. Knowledge of the prion structure will provide important insights into the self-propagation mechanisms of protein misfolding

    Heat shock protein amplification improves cerebellar myelination in the Npc1nih mouse model.

    BACKGROUND: Niemann-Pick disease type C (NPC) is a rare prematurely fatal lysosomal lipid storage disease with limited therapeutic options. The prominent neuropathological hallmarks include hypomyelination and cerebellar atrophy. We previously demonstrated the efficacy of recombinant human heat shock protein 70 (rhHSP70) in preclinical models of the disease. It reduced glycosphingolipid levels in the central nervous system (CNS), improving cerebellar myelination and improved behavioural phenotypes in Npc1nih (Npc1-/-) mice. Furthermore, treatment with arimoclomol, a well-characterised HSP amplifier, attenuated lysosomal storage in NPC patient fibroblasts and improved neurological symptoms in Npc1-/- mice. Taken together, these findings prompted the investigation of the effects of HSP amplification on CNS myelination. METHODS: We administered bimoclomol daily or rhHSP70 6 times per week to Npc1-/- (BALB/cNctr-Npc1m1N/J, also named Npc1nih) mice by intraperitoneal injection from P7 through P34 to investigate the impact on CNS myelination. The Src-kinase inhibitor saracatinib was administered with/without bimoclomol twice daily to explore the contribution of Fyn kinase to bimoclomol's effects. FINDINGS: Treatment with either bimoclomol or rhHSP70 improved myelination and increased the numbers of mature oligodendrocytes (OLs) as well as the ratio of active-to-inactive forms of phosphorylated Fyn kinase in the cerebellum of Npc1-/- mice. Additionally, treatment with bimoclomol preserved cerebellar weight, an effect that was abrogated when co-administered with saracatinib, an inhibitor of Fyn kinase. Bimoclomol-treated mice also exhibited increased numbers of immature OLs within the cortex. INTERPRETATION: These data increase our understanding of the mechanisms by which HSP70 regulates myelination and provide further support for the clinical development of HSP-amplifying therapies in the treatment of NPC.     RGS4 RNA secondary structure mediates Staufen2 RNP assembly in Neurons

    RNA‐binding proteins (RBPs) act as posttranscriptional regulators controlling the fate of target mRNAs. Unraveling how RNAs are recognized by RBPs and in turn are assembled into neuronal RNA granules is therefore key to understanding the underlying mechanism. While RNA sequence elements have been extensively characterized, the functional impact of RNA secondary structures is only recently being explored. Here, we show that Staufen2 binds complex, long‐ranged RNA hairpins in the 3′‐untranslated region (UTR) of its targets. These structures are involved in the assembly of Staufen2 into RNA granules. Furthermore, we provide direct evidence that a defined Rgs4 RNA duplex regulates Staufen2‐dependent RNA localization to distal dendrites. Importantly, disrupting the RNA hairpin impairs the observed effects. Finally, we show that these secondary structures differently affect protein expression in neurons. In conclusion, our data reveal the importance of RNA secondary structure in regulating RNA granule assembly, localization and even-tually translation. It is therefore tempting to speculate that secondary structures represent an important code for cells to control the intracellular fate of their mRNAs
